چکیده

Leucocyte alkaline phosphatase activity was determined in both cell homogenates and cell extracts. Leucocytes were obtained from normal controls, pregnant females, and patients with various haematological disorders. Enzyme activity presented in terms of phosphorus per minute per 10 neutrophils was more in agreement with the reported cytochemical data in both normal and pathological cases, than the values expressed in terms of phosphorus per minute per 10 leucocytes or per protein. This was explained by the possible low enzyme activity of lymphocytes included in the latter. Cases of acute and chronic granulocytic leukaemias showed very low activity compared to normal ̂ irrespective of the method of presentation of activity. Characterization of leucocyte alkaline phosphatase isoenzyme in normal subjects, using substrate specificities and selective inhibition tests, showed that it is largely similar to that of liver, bone, kidney and spleen, but differs markedly from those of the intestine and placenta. In this respect, the leucocyte alkaline phosphatase in pathological cases showed no variation from normal, except in cases of acute and chronic granulocytic leukaemias and pregnancy, where the leucocyte alkaline phosphatase enzyme showed a tendency to be heat resistant. In addition, the enzyme from acute gramdocytic leukaemia eases showed ä higher affinity for -naphthylphosphate, o-carboxyphenylphosphate, and adenosine S'-monophosphate.

The levels of Serum Alkaline Phosphatase and Lactate Dehydrogenase in Hodgkin Lymphoma

Background: Hodgkin’s disease (HD) is a neoplastic disease originating in lymphoid tissue, which spreads to lymphoid structures and ultimately nonlymphoid tissues. Lactate Dehydrogenase and Alkaline Phosphatase are increased in blood following membrane cell damage. The aim of this study was to compare Lactate Dehydrogenase and Alkaline Phosphatase levels in children in different stages of Hodg...

P-178: Separation and Identification of Alkaline Phosphatase Isozymes during Pregnancy

Background: Alkaline phosphatase (ALP), (EC 3.1.3.1) is a hydrolase enzyme responsible for removing phosphate groups from various molecules in the body. In humans, ALPis present in all tissues such as liver, bile duct, kidney, bone, and the placenta which detection of its activity is so useful in molecular biology. Pregnancy is associated with normal physiological changes that assist fetal surv...

Quantification and comparison of bone-specific alkaline phosphatase with two methods in normal and paget’s specimens

Background: Bone-specific alkaline phosphatase (BAP) is synthesized by the osteoblasts and is presumed to be involved in the calcification of bone matrix, though its precise role in the formation process is unknown. The aim of the present study was to measure the BAP activity in Paget's and normal specimens by two different techniques. Methods: Total ALP (TAP) as well as BAP activity-measuring ...

Immobilization of the Alkaline Phosphatase on Collagen Surface via Cross-Linking Method

Background: Collagen, the most abundant protein in the human body, and as an extracellular matrix protein, has an important role in the fiber formation. This feature of the collagen renders  establishment of the structural skeleton in tissues. Regarding specific features associated with the collagen, such as, formation of the porous structure, permeability and hydrophilicity, it can also be use...
